During the “Disa’s Plot" quest, you have to get inside Vernworth Castle, which is where knowing how to reach the Queen Regent's room in Dragon's Dogma 2 comes into its own.
While this Dragon's Dogma 2 mission is a fetch quest, finding the Queen Regent’s location in the castle is slightly confusing. There are multiple levels to the castle and several ways to get around, so you might be running in circles trying to reach the quest’s destination.

How to find the Queen Regent’s chambers in Dragon’s Dogma 2
When you arrive at Vernworth Castle, your Pawns have to stay behind, so it will just be you to navigate the grounds. You’ll have a guard who escorts you to a side entrance of the castle but from there, it’s all up to you.
After entering the side door, look on your map and find the large room that precedes the throne room. It has fountains in it and the room is located in the middle of the first floor of the castle.
When you find it, look for the spiral staircase that’s located on the south side of the room, as seen in the screenshots below. One of the screenshots shows the staircase’s location on the map while the other shows what the staircase looks like.
This is the staircase you need to climb to reach the second floor of the castle, which is where you’ll find the Queen Regent’s chambers and be able to reach your quest marker. Follow the hallways until you’re close to the yellow quest marker, which is inside the “Vernworth Castle - Guest Chambers” room. This isn’t technically where the Queen Regent resides but it’s where the evidence you’re looking for is, so head inside once you reach the door.
Inside, you can find a Torn Letter sitting on a desk, which is the evidence you need. Once you loot the Torn Letter, your quest log updates to say “You uncovered evidence of Disa’s scheme.” After seeing that, head out the open window in the room and leave the castle grounds.
Return to Captain Brant with the letter and you’ll learn more about what Queen Disa is planning with the False Sovran. You’ll also learn about her suspicious ties to another region in Dragon’s Dogma 2.
